Rei Saotome (早乙女レイ Saotome Rei) is a young girl whose talent at dueling manages to surpass the expectancy for her age. She sneaks into Duel Academia disguised as a boy, and by her return to the Academia she wins the right to become an Osiris Red student. Rei's love interest seems to shift multiple times, until she finally came on the decision of Judai Yuki.

At the Academia, she meets Martin Kanou who becomes her friend.
After the Academia is warped through space by Yubel, Rei asks Martin to go with her to the restroom, but Yubel attacks her midway. She gets really sick and is need of medical help. Judai and the others go through the desert trying to find the medicine, while Rei's fever gets worse.

Halfway through this arc, Rei was promoted to Obelisk Blue but still wears her Osiris Red outfit.

Her second deck was a Mystic deck, which she used to display her ability at the GX Tournament while facing Jun Manjoume. Although she ended up losing, many people at the Academy admired her skill, and decided to let her in the academy even though she was just 12.
